How depression and antidepressant drugs work: New insight from depressed mice helps researchers unite two hypotheses

Mutations in the GABA-A receptor cause a temporary imbalance between GABA signaling (yellow) and glutamate signaling (blue) in the brain. Homeostatic down regulation of glutamate signaling rebalances the system at a lower level and can lead to depression. Treatment with ketamine restores both GABA and glutamate levels to normal. Structure of the RNA-modifying protein RlmN, shown in a ribbon-diagram. The RlmN (blue ribbon) is trapped in the middle of its reaction while it is bound to transfer RNA (shown in grey, stick format). Iron and sulfur atoms are shown as orange and yellow spheres. Selected amino acids, cofactors, and nucleobases are shown in stick format and are colored by atom type. CREDIT: Penn State
